Effects of Bupleuri radix aqua - acupuncture on antioxidative enzymes and lipid peroxidation induced by acetaminophen in mouse


Abstract
The drug induced liver necrosis has been extensively studied in mice and rats using acetaminophen intoxication as a model system. Lipid peroxidation has been discussed as a basic deteriorative mechanism which leads to liver dysfunction. In the previous study, we reported that Bupleuri radix aqua-acupuncture solution(BRAS) exhibited inhibitory effect on linoleic acid peroxidation, also exhibited inhibitory effect on superoxide generation, and showed scavenging effect on DPPH radical. Furthemore, BRAS protected the cell death induced by t-BHP in the normal rat liver celi dose-dependentty. To further understand these effects of BRAS, this study was purposed to investigate the antioxidative effect of BRAS on acetaminophen induced lipid peroxidation in mice. We administerated acetaminophen(500§·/§¸, i.p.) to starved ICR mice for 24hr after pretreatment of BRAS for 6days. In the liver mitochondrial and cytosolic frations, amounts of lipid peroxide and glutathione(GSH), and activities of hepatic enzymes were measured. In vivo administeration of BRAS showed signifcant inhibition of acetaminophen induced lipid peroxidadon and GSH depletion, and elevations of superoxide dismutase activity. These results suggested that BRAS might play a protective role in lipid peroxidation by free radicals in vivo.
